Peripheral Blood CD34+ Cells Differ From Bone Marrow CD34+ Cells in Thy-l Expression and Cell Cycle Status in Nonhuman Primates Mobilized or Not Mobilized With Granulocyte Colony-Stimulating Factor and/or Stem Cell Factor

چکیده
Granulocyte colony-stimulating factor (G-CSF) and stem cell factor (SCF) have been shown t o stimulate the circulation of hematopoietic progenitor cells in both mice and nonhuman primates. We evaluated the immunophenotype and cell cycle status of CD34+ cells isolated from the bone marrow (BM) and leukapheresis product of cytokine-mobilized nonhuman primates. CD34+ cells were isolated from rhesus macaques that had received no cytokine therapy, 100 pg/kg/d G-CSF, 200 pg lkg ld SCF, or a combination of both 100 pglkgld GCSF and 200 pglkgld SCF as a subcutaneous injection for 5 days. BM was aspirated before (day 0) and on the last day (day 5) of cytokine administration. On days 4 and 5, peripheral blood IPB) mononuclear cells were collected using a novel method of leukapheresis. Threefold more PB mononuclear cells were collected from animals receiving G-CSF alone or G-CSF and SCF than from animals that had received either SCF alone or no cytokine therapy. CD34+ cells were positively selected using an immunoadsorptive system from the BM, PB, andlor leukapheresis product. Threefold and 10fold more CD34' cells were isolated from the leukapheresis product of animals receiving G-CSF or G-CSF and SCF, respectively, than from animals receiving no cytokine therapy

متن کاملA Comparative Study of the Phenotype and Proliferative Capacity of Peripheral Blood (PB) CD34+ Cells Mobilized by Four Different Protocols and Those of Steady-Phase PB and Bone Marrow
Peripheral blood (PB) CD34+ cells from four commonly used mobilization protocols were studied to compare their phenotype and proliferative capacity with steady-state PB or bone marrow (BM) CD34' cells. Mobilized PB CD34+ cells were collected during hematopoietic recovery after myelosuppressive chemotherapy with or without granulocytemacrophage colony-stimulating factor (GM-CSF) or granulocyte c...

Gene transfer experiments in nonhuman primates have been shown to be predictive of success in human clinical gene therapy trials. In most nonhuman primate studies, hematopoietic stem cells (HSCs) collected from the peripheral blood or bone marrow after administration of granulocyte colony-stimulating factor (G-CSF) + stem cell factor (SCF) have been used as targets, but this cytokine combinatio...
متن کاملCD34 expression by murine hematopoietic stem cells mobilized by granulocyte colony-stimulating factor.
Controversy has existed about CD34 expression by hematopoietic stem cells. We recently reported that CD34 expression reflects the activation state of stem cells by using a murine transplantation model. It has been generally held that mobilized blood stem cells express CD34.However, it has also been reported that mobilized stem cells and progenitors are in G0/G1 phases of the cell cycle. To addr...

Peripheral blood stem cells (PBSCs) mobilized with high-dose chemotherapy and hematopoietic growth factors are now widely used to support myeloablative therapy of multiple myeloma and effect complete remissions in up to 50% of patients with apparent extension of event-free and overall survival. Because tumor cells are present not only in bone marrow, but also in virtually all PBSC harvests, it ...
